SLU-PP-332 is a synthetic estrogen-related receptor (ERR) α/β/γ agonist being investigated as a potential exercise-mimetic compound. ERRs are nuclear receptors involved in regulating mitochondrial function, energy metabolism, fatty-acid utilization, and skeletal-muscle adaptation.
Research on SLU-PP-332 has generated significant interest because activation of the ERR pathway can produce molecular and metabolic changes associated with aerobic exercise.
Preclinical studies found that SLU-PP-332 increased mitochondrial respiration and oxidative muscle-fiber characteristics and improved exercise endurance in mice.
Additional mouse research found increased energy expenditure and fatty-acid oxidation, along with reductions in fat-mass accumulation and improvements in insulin sensitivity in models of obesity and metabolic syndrome.
SLU-PP-332 activates all three ERR subtypes — ERRα, ERRβ, and ERRγ — with the greatest activity toward ERRα. These receptors function as important regulators of genes involved in mitochondrial metabolism and the physiological response to exercise.
Rather than acting as a traditional stimulant, SLU-PP-332 is being investigated for its ability to influence the cellular metabolic machinery associated with exercise adaptation.
