Mazdutide is a long-acting dual agonist of the GLP-1 and glucagon receptors being investigated for its effects on body weight, appetite regulation, glucose metabolism, energy balance, and metabolic health.
Unlike compounds that target GLP-1 alone, mazdutide activates both GLP-1 and glucagon signaling, creating a broader metabolic profile that researchers are studying for weight management and cardiometabolic applications.
GLP-1 signaling is associated with appetite regulation, glucose-dependent insulin secretion, and delayed gastric emptying.
Glucagon signaling plays an important role in energy expenditure, glucose regulation, and mobilization of stored energy.
Together, these pathways are being investigated for their potential effects on:
Mazdutide has progressed through Phase 3 clinical development. In a 48-week Phase 3 trial, once-weekly 4 mg and 6 mg doses produced mean body-weight reductions of 11.0% and 14.0%, respectively, compared with 0.3% with placebo. Improvements were also observed across prespecified cardiometabolic measures.
Higher-dose research has also generated significant interest. In a Phase 1 study using doses escalated up to 16 mg, participants receiving mazdutide experienced approximately 20–21% mean weight reduction at week 20; gastrointestinal effects were the most commonly reported adverse events.
Mazdutide represents a different approach to metabolic signaling by combining:
GLP-1 Activity + Glucagon Activity → Appetite Regulation + Metabolic Signaling + Energy Balance
This makes it particularly interesting for research involving obesity, metabolic dysfunction, body composition, glucose regulation, and energy metabolism.
Clinical studies have most commonly reported gastrointestinal adverse events such as nausea, diarrhea, decreased appetite, and related GI symptoms, generally ranging from mild to moderate.
Research findings from clinical trials should not be interpreted as establishing an appropriate dose or protocol for an individual.
