ARA-290, also known as cibinetide, is an 11-amino-acid peptide derived from the three-dimensional structure of erythropoietin (EPO). Unlike EPO, ARA-290 was designed to activate the innate repair receptor (IRR) without stimulating erythropoiesis, making it an important compound in research surrounding tissue protection, inflammation, and nerve repair.

ARA-290 is being investigated for its ability to activate signaling pathways associated with anti-inflammatory responses, tissue protection, neuronal repair, and cellular survival.

Research suggests the innate repair receptor becomes more active following tissue or nerve injury. ARA-290 may help shift the local environment away from inflammatory and tissue-damaging signaling toward processes associated with repair and recovery.

Neuroregenerative Research

One of the most interesting areas of ARA-290 research is small-fiber neuropathy and nerve regeneration.

In clinical studies involving patients with sarcoidosis-associated small-fiber neuropathy, ARA-290 was associated with improvements in neuropathic and autonomic symptoms, sensory function, quality of life, and measures of corneal nerve fiber density.

A separate Phase 2 study in people with type 2 diabetes and painful neuropathy reported improvements in neuropathic symptoms and certain metabolic measures following 28 days of treatment.

Tissue Protection & Inflammation

Preclinical research has also investigated ARA-290 in models of nerve injury and inflammatory disease. In animal models, activation of the innate repair receptor was associated with reduced neuroinflammatory signaling, improved nerve regeneration, and tissue-protective effects.
