What Reconstitution Means

Reconstitution is the process of adding a liquid diluent to a lyophilized (freeze-dried) peptide to return it to a solution that can be used in an experimental protocol. As covered in the first lesson, peptides ship freeze-dried because they’re unstable in solution — reconstitution is the step that reverses that, and it’s meant to happen immediately before the material is needed, not far in advance.

Why Bacteriostatic Water Specifically

Bacteriostatic water is water for injection that contains a small amount (typically 0.9%) of benzyl alcohol as a preservative. That preservative inhibits bacterial growth in the solution after reconstitution, which is why it’s the standard diluent choice in this space rather than plain sterile water — it extends how long a reconstituted vial stays usable before contamination becomes a concern.

Variables That Affect Stability

  • Agitation: diluent is typically added gently down the inside wall of the vial rather than injected directly onto the powder or shaken forcefully — vigorous agitation can cause some peptides to denature or aggregate
  • Temperature: reconstituted solutions are generally kept refrigerated, not at room temperature, to slow degradation
  • Light exposure: some compounds are light-sensitive and are stored in amber vials or foil-wrapped for this reason
  • Time: even under good storage conditions, reconstituted peptides have a finite usable window before potency declines meaningfully
